Locations have associated difficulties for terrain (easy - impossible) and level of shadow (free lands - dark lands).
RAW are the associated Difficulty Levels applied to Travel and Corruption tests in those locations, or do they just affect the frequency / likely hood of those tests?

For fatigue tests, use a target number of 14, unless you want to use the difficulty of the region. If you do, use the difficulty of the region on the longest leg of the journey, or break up rolls into difficulty per region. Roll as many fatigue tests as called for by the rules for journeys.
Use corruption tests to resist corruption from blighted places. The frequency is determined by the type of region on the Blighted Lands table, and it only applies when characters are in an area the Loremaster considers a blighted place. Use the default target number of 14 unless you want to use the difficulty of the region.

Personally, I always use the difficulty of the particular Region, as it makes it a bit more challenging.
Adventure Summaries for my long-running group (currently playing through The Darkening of Mirkwood/Mirkwood Campaign), and the Tale of Years for a second, lower-level group (in the same campaign).

Me too.
I use the Blighted Lands table (page 223) in the Core Rulebook for the frequency of blighted lands occurring. The frequency of Fatigue Tests are determined by the season (page 158). I use the Region Table (page 158) for the TN.

For Fatigue Tests I use the terrain type (eg, Easy, Medium, Hard, etc) and for Corruption Tests the region type (eg, Free Lands, Border Lands, Shadow Lands, etc). For other types of Hazards I play a bit looser and consider the type of Hazard, then deciding on whether to use the region type or the terrain type. This approach give me the flexibility I need and has worked well during my campaign.

Yet another combination :
For Fatigue and Hazard test, I always use the Terrain difficulty.
For Corruption tests, I consider how "powerful" the blighted place is and adjust the TN acordingly. The Terrain difficulty is a guide, but might not be appropiate for the corruption I'm presenting. For instance, Smaug's remnants are probably much more difficult to resist than the usual TN for Lake-town's surroundings.
